后毒副反应小,因此用M中作为肿瘤的过继免疫治疗值得进一步探讨.本实验的研究目的:探索MΦ过继免疫治疗对实验动物移植性肿 瘤的抑制效应。观察抗原冲击的M中瘤苗能否通过激发淋巴细胞的特 异性免疫而增强其抑瘤效应。为此主要进行如下工作:1.体外活化M中及制备抗原冲击的M中瘤苗(Ag-pulsedM中):提取 BALB/C鼠腹腔MΦ,体外以1000u/mlGM-CSF和200w/mlIFN-Y刺 激48小时,然后加入500ug/ml的艾氏腹水瘤或S180肿瘤的细胞膜 碎片(冻融法或超声粉碎法制备,作为肿瘤细胞相关抗原)孵育2小 时,消化收集,即得Ag-pulsedMΦ.2.
